RESUMO

Cryopreservation of small ruminant's semen is an effective strategy for distributing spermatozoa for reproductive programs, but this process decreases the fertility potential of post-thawed spermatozoa. The aim of this research was to assess the effect of different concentrations of CoQ10 in soybean lecithin (SL)-based extender on buck semen quality during cryopreservation process. Semen samples were collected from five bucks, twice a week, then diluted in the SL-based extender containing different concentrations of CoQ10 as follows: extender containing 0 µM (control, Q0), 0.1 µM (Q0.1), 1 µM (Q1), 10 µM (Q10) and 100 µM (Q100) CoQ10. Motion characteristics, membrane functionality, abnormal morphology, mitochondrial activity, acrosome integrity, viability, apoptotic-like changes, lipid peroxidation, DNA fragmentation and ROS concentration were evaluated after freeze-thawing process. The Q10 resulted in greater (P≤0.05) total motility, progressive motility, average path velocity, membrane integrity, mitochondrial activity, acrosome integrity and viability compared to the other groups. Furthermore, supplementation of freezing extender with 10 µM of CoQ10 presented lower (P≤0.05) apoptotic-like changes, lipid peroxidation, DNA fragmentation and ROS concentration compared to the other groups. Regarding to the protective effect of CoQ10 supplement during cryopreservation process, it could be explored as a potent antioxidant for cryopreservation of buck semen as it preserved the post-thawed buck sperm quality.

Sperm cryopreservation is one of the main methods for preserving rooster sperm for artificial insemination (AI) in commercial flocks. Yet, rooster sperm is extremely susceptible to reactive oxygen species (ROS) produced during the freezing process. Oxidative stress could be prevented by using nanoparticles containing antioxidants. The present study was conducted to investigate the effect of zinc oxide nanoparticles (ZnONP) in rooster semen freezing extender on quality parameters and fertility potential. For this aim, semen samples were collected and diluted in Lake extenders as follows: control: Lake without ZnONP, ZnO100: Lake with 100-µg zinc oxide (ZnO), ZnONP50: Lake with 50-µg ZnONP, ZnONP100: Lake with 100-µg ZnONP and ZnONP200: Lake with 200-µg ZnONP. After freezing and thawing, sperm motility, viability, membrane integrity, morphology, mitochondrial activity, acrosome integrity, DNA fragmentation, lipid peroxidation and ROS, as well as fertility and hatchability were assessed. According to the current results, higher rates of motility, membrane integrity, mitochondrial activity, acrosome integrity and live cells were detected in the ZnO100, ZnONP50 and ZnONP100 groups compared to other groups (p ≤ .05). Yet, the percentage of dead cells, DNA fragmentation, lipid peroxidation and ROS levels were lower in the mentioned groups (p ≤ .05). Furthermore, a higher percentage of fertility was observed in the ZnO100 and ZnONP100 groups than in the control group (p ≤ .05). In conclusion, the use of 100-µg ZnO and 50- to 100-µg ZnONP represents a valuable and safe additive material that could be used to improve the quality and fertility potential of rooster sperm under cryopreservation conditions.

Broilers under oxidative stress from high ambient temperatures may benefit from the use of additives that have antioxidant properties. This experiment investigated the efficacy of a herbal extract mixture (HEM; aqueous extracts from Ferula gummosa, Thymus vulgaris, and Trachyspermum copticum) in day-old chicks, injected intramuscular (deep pectoral muscle; (0, 30, 60, and 90 µL/0.1 mL of sterilized and distilled water)), and supplemented in drinking water (0 and 0.25 mL/L) during the rearing period. Broilers were reared in battery cages under summer temperature conditions, with average maximum temperature of 35.5°C, average minimum temperature of 25.5°C, and average relative humidity of 50-60%. A total of 400 1-day-old Ross 308 male broiler chicks were randomly assigned to 8 treatment groups (5 replicates/treatment with 10 birds per replicate). From d1 to d10, the indoor air temperature was adjusted to match fluctuating outdoor summer temperatures, and was set at 30-34°C and 50-60% relative humidity; and from d10 onwards, no adjustments were made. Injection of HEM linearly decreased feed:gain (P = 0.005), heterophile-to-lymphocyte (H/L) ratio (P = 0.007), and serum concentrations of cholesterol (P = 0.008), low-density lipoprotein cholesterol (LDL) (P < 0.001), malondialdehyde (P = 0.005), and cortisol (P = 0.008). The 60 µL of HEM injection produced the best results in terms of final body weight (BW; P = 0.003), overall average daily gain (ADG; P = 0.002), European performance index (P < 0.001), carcass yield (P < 0.001), and serum glutathione peroxidase activity (P < 0.001). Supplementation of HEM in drinking water also increased final BW (P = 0.048), overall ADG (P = 0.047), high-density lipoprotein cholesterol (P = 0.042), and total antioxidant capacity (P = 0.030), while decreasing the H/L ratio (P = 0.004) and serum LDL concentration (P = 0.031). There were interactions between injection and water supplementation for BW (day 24; P = 0.045), carcass yield (day 42; P = 0.014), and serum superoxide dismutase activity (day 42; P = 0.004). In conclusion, administering an injection of HEM at a dose of 60 µL at the time of hatching, followed by supplementation at a dose of 0.25 mL/L via drinking water during the rearing period could be a useful strategy for improving the performance and health status of heat-stressed broiler chickens.

The effects of a grain source (corn grain (CG) vs. barley grain (BG)) and starter protein content (19% vs. 22% CP, dry matter basis) on growth performance, digestibility, ruminal fermentation, and blood metabolites were evaluated in Holstein dairy calves. Forty 3-day-old female calves with a starting body weight of 39.3 kg were subjected to four treatments in a completely randomized design with two by two factorial arrangements. Treatments were: (1) CG + 19% CP (CG-19CP); (2) CG + 22% CP (CG-22CP); (3) BG + 19% CP (BG-19CP); and (4) BG + 22% CP (BG-22CP). All calves were weaned at 59 days of age and remained in the study until 73 days of age. Starter and total DM intake were not affected by grain source and dietary protein content (p > 0.05). The average daily gain and feed efficiency were improved, and ruminal total short-chain fatty acid, propionate, and butyrate concentrations were increased in BG calves compared to CG calves (p < 0.05). The ruminal concentrations of ammonia nitrogen (d 71; p = 0.02) and acetate (d 35; p = 0.02) were increased in CG fed calves compared to BG. The greatest wither height (p = 0.03) and blood insulin concentration (p = 0.03) were seen in BG-22CP treatment. In conclusion, BG has marginal benefit in the height of calves when fed with diet containing 22% CP which may be recommendable in replacement heifer rearing programs.

This study was conducted to determine the optimum level of home-made selenium-enriched yeast (SeY) in the diet of broiler breeder hens and to compare the effects of this product with sodium selenite (SS) or Selemax (SM) on their productive and reproductive performance. A total of 150 broiler breeder hens were divided to six groups and hens in each group were received a basal diet containing no selenium (CG), 0.15, 0.30, 0.45 mg SeY/kg diet (SeY-0.15, SeY-0.30 and SeY-0.45, respectively), 0.30 mg SM/kg diet or 0.30 mg SS/kg diet for 15 successive weeks. The results showed that egg weight and production and hatchability rate were higher in SeY-0.45 compared to other groups (P < 0.05). Also, SeY-0.45 group led to lower embryonic mortality rate compared to CG and SS groups. Fertility rate and chick quality parameters were not affected by selenium supplementation during this period (P > 0.05). In conclusion, the dietary supplementation of home-made selenium, as an organic selenium source, can be used to improve the productive and reproductive performance in aged broiler breeder hens at 0.45 mg/kg feed.

The effects of coenzyme Q10 (CoQ10) has not yet been assessed for cryopreservation of rooster semen. The aim of this study was to evaluate the effect of different concentrations of CoQ10 in Lake extender for cryopreservation of rooster semen. The viability and apoptosis status, DNA fragmentation, abnormal morphology, motion parameters, membrane functionality, mitochondrial activity, acrosome integrity, lipid peroxidation, and fertility potential were evaluated after the freeze-thaw process. Semen samples were collected from ten roosters, twice a week, and then diluted in extender contained different concentrations of CoQ10 as follows: Lake without CoQ10 (control, Q 0), Lake containing 1 µM (Q 1), 2 µM (Q 2), 5 µM (Q 5), and 10 µM (Q 10) CoQ10. Supplementation of Lake with 1 and 2 µM CoQ10 resulted in greater sperm viability, total motility, progressive motility, membrane functionality, mitochondrial activity, acrosome integrity, and fertility rate. Furthermore, the extent of lipid peroxidation in thawed spermatozoa treated with 1 and 2 µM CoQ10 was less than with the other groups. Different concentrations of CoQ10 had no effect on DNA fragmentation and sperm morphology. Results of the present study indicate that supplementation of Lake extender with 1 and 2 µM CoQ10 enhances the quality of rooster sperm after the freeze-thaw process.

A two-month trial was conducted to investigate the effects of pelleting rations with 70:30, 65:35, and 60:40 concentrate to high-quality roughage (alfalfa hay) ratios on hematological and biochemical parameters of ostriches. A total of 18 seven-month-old male ostrich chicks, initially weighing an average of 60-70 kg, were distributed into three different outdoor paddocks at a stocking density of six birds per paddock. In the morning, blood collection was made from the wing vein after about 12 h of fasting. All rations resulted in no significant change in hematological parameters. However, with increasing alfalfa inclusion rate, the heterophil/lymphocyte ratio tended to be lower (P = 0.089); whereas the values of hemoglobin, packed cell volume (PCV), and mean corpuscular hemoglobin (MCH) tended to be higher (P values 0.069, 0.072 and 0.094, respectively). In terms of plasma biochemical parameters, increasing the ratio of concentrate to alfalfa hay in ostrich diets resulted in significant (P < 0.05) depression in the values of glucose (up to 20.2 %), total cholesterol (up to 19.2 %), creatine kinase (up to 27.9 %), and aspartate aminotransferase (up to 29.9 %). Additionally, the 65:35 or 60:40 rations caused a decrease in low-density lipoprotein cholesterol level (27.9 and 42 %, respectively; P = 0.008) compared to the 70:40 ration. Without exception, no rations affected the values of leukocyte subsets, total protein, albumin, globulin, uric acid, alanine aminotransferase, gamma glutamyl transferase, triglycerides, high-density lipoprotein cholesterol, and very low density lipoprotein. Based on our results, the concentrate to alfalfa ratio of 60:40 could be suggested as optimum ratio for good health conditions of juvenile ostriches.

The objective of this study was to examine the interaction of different concentrations of trehalose [0 (T0), 50 (T50) or 100 (T100) mM] and glycerol [5% (G5) or 7% (G7)] on post-thawed quality of ram semen, cryopreserved in a soybean lecithin (SL)-based extender. Twenty-eight ejaculates were collected from four rams and diluted with six trehalose/glycerol combinations: T0G5, T50G5, T100G5, T0G7, T50G7, and T100G7. Sperm motility (CASA), membrane integrity (eosin/nigrosin) and functionality (HOST), abnormal forms, capacitation status (CTC), mitochondrial activity (rhodamine 123), apoptotic features (Annexin V/propidium iodide) and lipoperoxidation (malondialdehyde production) were evaluated after thawing. Extender T100G5 yielded the highest results for total and progressive motility, sperm velocity, normal morphology, functional membranes, active mitochondria and membrane integrity, with P<0.05 in general, except for T50G7 (P>0.05). The combinations T0G5, T0G7 and T100G7 yielded the lowest post-thaw quality. We could not detect significant changes in other kinematic parameters, capacitation status or lipoperoxidation. We conclude that, in our SL-based extender, a combination of 100 mM trehalose and 5% glycerol was the most adequate combination to achieving post-thawing quality in our soybean lecithin-based extender, and our results support that a synergistic effect among trehalose and glycerol exists. We suggest that other combinations could improve these results.
